As of 2012, the quinolone antibacterials that have been developed and put into mass production in China are mainly norfloxacin, ciprofloxacin, ofloxacin, enoxacin, lomefloxacin, and fleroxacin. Among them, norfloxacin, ciprofloxacin and ofloxacin produced the largest amount, accounting for about 98% of the total domestic production of fluoroquinolones.
Fluoroquinolones are generally obtained by the synthesis of a fluoroquinoline-containing compound from a fluorinated benzene ring followed by condensation with piperazine (or methylpiperazine). Due to the abundant reserves of fluorite in China, it is one of the countries with the largest production of fluorine-containing drugs and intermediates in the world, and exports more than 80% of fluorine-containing intermediates. As a whole, China's fluorobenzene intermediates developed earlier and the production capacity was generally excessive; the development of trifluorotoluene intermediates was relatively late and the development speed was fast; while for heterocyclic aromatic compounds, especially fluoropyridines, China only had Some research units and manufacturers own the synthesis technology of fluoropyridine intermediates. Therefore, the fluoropyridine intermediates will become one of the main directions for the research and development of fluorine intermediates in the next few years.
